Abstract:In order to identify the resistance gene (s) to strip rust in M97, F1, F2, F3 and BC1 progenies, derived from M97 crossing with susceptible parent Mingxian169, were tested with Chinese races Sun11-4, Sun11-11, CY29, CY30, CY31, CY32 and CY33 at the seedling stage in greenhouse. The gene conferring resistance to Sun11-4 in M97 was mapped by linked SSR markers. The genetic results showed that one dominant gene conferring resistance to Sun11-4 and Sun11-11; one dominant gene and one recessive gene conferring resistance to races CY29, CY30 and CY33; two dominant genes independently conferring resistance to CY31. The mapping population was constructed with F2 generation inoculated with Sun11-4. Four SSR markers, Xwmc222, Xwmc147, Xbarc229 and Xwmc339, were link to the resistance gene, with genetic distance range from 3.9 to 11.2 cM. The gene was located at chromosome 1DS and the gene maybe a new gene different from all known stripe rust resistance genes, temporarily designated as YrM97. Flanking markers of Xwmc222 and Xwmc147 were used to test 42 wheat cultivars from Huanghuai wheat growing region, only 9.5% cultivars have the same polymorphic bands as YrM97, which indicated the polymorphic SSR markers can be useful in wheat molecular marker select breeding.
