犃犫狊狋狉犪犮狋:Inthisstudy,aF1populationofalfalfawasgeneratedbycrossinganearlymaturing,lowyield
plant(maleparent)andalatematuring,highyieldingplant(femaleparent).AndasystemofPCRampli
ficationfortetraploidalfalfawasestablishedbyanalyzingthestandardPCRreactionprogramortouch
downprogram.Totalof130SSRmarkersweretestedintheexperiments,ofwhich60markersgavepoly
morphicbetweentwoparentsandwereusedtoconstructthegeneticlinkagemap.GenotypesoftheF1
mappingpopulationwereanalyzedbyPAGEelectrophoresis.Andaprelimarygeneticlinkagemapwas
constructedbyusingTetraploidMapsoftware.Thegeneticmapconsistsof44loci,spanning979cMand
coveringalof8alfalfachromosomeswithanaveragegeneticdistanceof22.25cM.Moremarkersarestil
neededtobeaddedtomakeasaturatedgeneticmap.Thestudymadeafoundationforidentifyingimpor
tantquantitativetraitloci(QTL)forimportantagriculturaltraits.
