Abstract:There was a wide range of variation (from 1.04% to 14.21%) among 12 genotypes for callus induction frequency, although all of them formed calli. Cultivars, lines and F1 hybrids carring BYMV resistance genes had higher callus induction frequency (8.83%-14.21%) than that of others (1.04%-6.25%). The comparison of callus induction frequency in four media revealed that when the medium was supplemented with 1.0 mg/L 2, d-D, 0.1 mg/L Kt, 0.2 mg/L BA and 0.1 mg/L Biotin, it was more suitable (ll.09%)than the other three media (6.33 %, 8.21% and 10.82% respectively). The results indicated that after incubating the anther at 15℃ for 5 days, then incubate them at 25℃ in the dark(condition 1), increased the callus induction frequency (from 5.84% to 11.59%), although the peak of the callus induction appeared 2-4 days later. No statistical differences were found for the callus induction frequency between dark culture (condition 2) and light/dark culture (condition 3).
