05 一叶荻及一叶裁碱片中一叶荻碱测 定 方 法 的研

一叶获碱 ( S e o u r i in n e) 系自大戟科植物 一 叶
簌 〔S o e u r i n e g a s u f f r u t i e o s a ( p a l l . ) R e h d . 〕
叶中分离得的一种生物碱 。 具有明显脊髓兴奋作用 。 曾
用于多种神经疾病 。 为了设计一简便和准确的测定方
法 , 我们研究了用氧化铝薄层分离一叶荻提 取物中一
叶荻碱后 , 与澳甲酚绿生成有色离子对 , 用以进行比
色的方法 。 一叶荻碱浓度 ( 1 5 0卜g 以 内 ) 与 吸 收 度
呈良好线性 。
生药的测定 : 精密称取一叶荻叶 40 目粉 19 , 加
2 5纬氯氧化钱浸润后 , 精密加氧仿 l om l , 放丑过夜 ,

取氛仿提取液 4 ml, 水浴上蒸干 , 加无水乙醇定容至

, 点加此液 4 0~ 5 0林 1 (相当 1 6~ Z om g生药 ) 于
不含粘合剂氧化铝薄层上 , 用不含醇氛仿为展开剂 ,
分离一叶荻碱 , 趁薄层未千 , 于 2 5 4 n rn 紫 外 光 下 观
察 , 一叶荻碱呈黄色荧光 , 收集该斑点 , 用氯仿洗脱 ,
回收溶剂后 , 于残渣中加蒸馏水 s m l , 。 . 06 % 澳甲酚
绿溶液 ( P H 4 . 9 4 ) 2 m l , 氛仿 1 0 m l , 振摇 , 避光静
笠 , 分取澄 明纵仿液 6 m l , 加 O . 0 7 N氢氧化钾无水乙
醇溶液 工 m l , 于 6 2On m测吸收度 。 并取薄层上空白氧
化铝经同法处理后作空白校正 。
硝酸一叶荻碱片的测定 : 取10 片 , 精密称定 。 研
细 、 精密称出适量 (约相当硝酸一叶簌碱 Z m g) , 加
水振摇并定容至 50 m l , 用干燥滤纸过滤弃去初滤液 ,
取续滤液 2 m l , 加水 3 m l后 , 自加澳甲酚绿试液起同
上法处理测吸收度 , 并与苏联药典 ( X版 ) 测定方法
比较 , 所得结果相符 , 但该药典法消耗样品量较本法
约多 10 倍 。
本文研究了薄层层离条件与比色测定条件 , 方法
简便 、 不需特殊仪器 , 所用样 钻量少 , 结果稳定 , 回
收率为 9 . 0 土 2 . 3% 。
