Direct Extraction of Genomic DNA from Dry Micially-aged
Seeds of Flowering Chinese Cabbge and
its Application in RAPD Analysis
QUO ~ i - ~ i n @ FU Jia-Rui
( L k p r b n d 4Bidogionl Scienm and Bioiechndog.r, Zhangshan Unimisy, Guangzhou 510275)
Abstract Flowering Chinese cabbage seeds of diierent vigor were obtained by treatment at 40 t
and 100% RH for different days. Genomic DNA was extmcted with balanced phenol and chlorofoxm
directly from the dry aged seed. RAPD amplification was carried out on the extracted genomic DNA.
The extracted genomic DNA samples were large in quantity and identical. DNA bands amplified by
primer S208 were clear and bright. It could be concluded that genomic DNA extracted directly h m
the dry d ~ c i a l aged seeds of flowering Chinese cabbage could be used for RAPD analysis.
Key words Flowering Chinese cabbage, Dry seed, Artificial accelerated ageing, Genomic DNA,
