Abstract:Chromosome analysis and representational difference analysis were conducted on susceptible strain and disultap-resistance and deltamethrin-resistance and cartap-resistance near isogenic strain. The results showed that the number of chromosomal in susceptible and insecticide resistance strain was 2n=30 and the chromosomes present cosh-like. No distinct difference occurred between susceptible and resistance strain. Representational difference analysis (RDA) was applied to find more differences between susceptible and the other three resistant strains. Susceptible strain was used as driver and the other three as testers. Subtractive hybridization and dynamic enrichment were carried out and produced several different fragments. Through southern blot with partial probes we have got 2, 2 and 3 special fragments in the NILS of disultap-resistance, deltmethrin-resistance and cartap-resistance respectively. Compared with the susceptible strain, the resistant strains have no obvious changes in karyotype but have distinct difference in DNA level, which suggests that insecticide resistance of diamondback moth is relative to the genomic DNA changes.
