元宝枫及同属植物种子总蛋白质
含量和氨基酸的测定

摘 要:对元宝枫及同属植物种子总蛋白质含量和氨基酸的组成进行了分析测定 ,结果表明元宝枫
及同属植物种子含有较高的蛋白质 ,且其氨基酸的组成以谷氨酸 、精氨酸含量较高为特点.
关键词:元宝枫;种子;蛋白质;氨基酸

Abstract:An analy sis and determinat ion are made of the contents of the total protein and the
components of 19 kinds of amino acids in the Acer truncatum Bunge.And the same generic plants are
analyzed.The results show that there is a very high pro tein content in the seeds of Acer truncatum
Bunge and the same generic plants.Moreover , the higher contents of Glu.and Arg.is a peculiari ty of
the components of amino acids in the seeds of Acer truncatum Bunge and the same generic plants.

Acer truncatum Bunge.belong to the Acer
plant.Acco rding to folk record the cover bar be used
as medicine , according to dispelling the eff iciency of
the rheumat ism , Its tender leaf can take the place of
tea-drinking or make vegetables , the f rui t clears aw ay
heat , favo rable throat.In the no rtheast of our
country , the ex tensive application of the province ,
such as the Yellow River and the Yang ze River
v alley[ 1 ~ 2] , etc.The seed has only protein but not
strainer[ 3] .Seed oil fine edible oil and indust rial
oil[ 4~ 5] .In folk w hen the fruit has not been ripe , the
child likes to eat thei r fruits.A .derv idii Fr.
(QZHQ), A.max imowizcii Pax (WJQ), A.
tetramerum Pax.(SRQ), A.robustum Pax
(CHRQ), Acer cappadocium Rehd(QPQ), A.
erianthum Schwer.(MHQ), A.buergeriana Pax
(SJQ)belong to the same generic plants of Acer[ 6] .
With the same generic plants distribute ex tensive in
Shaanxi. To develop and utilize Shaanxi Acer
resources synthetically , we analyzed thei r seed of

protein and amino acid content.The results of
determined as follows.
1 Materials
(1)The seed of Acer truncatum Bunge.Is
collected Xi′an in December , October , and
September 2001.
(2)The seed of the same generic plants:A .
dervidii Fr.(QZHQ), A .max imowizcii Pax
(WJQ) , A .tetramerum Pax.(SRQ) , Acer
cappadocium Rehd(QPQ) and A .robustum Pax
(CHRQ)are collected f rom HuoDiTang of Ningshan
county;A .erianthum Schw er.(MHQ)is collected
from Hu county ;And A .buergeriana Pax(SJQ)is
collected Northwest forest ry institute.Professo r
Wang Ming-chang of northwest forest ry insti tute
appraised all samples.
(3)Reagent:Dense sulphuric acid;catalyst is
mixture of the sodium copper and sulphate sulphuric
acid;30% NaOH , standard hydrochloric acid (0.
0616N);mix indicator (methy l red , cresol green
bromine).All reagents is pure to analyses and
produced of the chemical reagent factory of Xi′an.
2 Methods
2.1 Treatment of the sample
To weigh certain detailed sample (deg rease), to
enter a certain measuring cup , put it drying 4 hours
in 105°of ovens , then put the measuring cup into the
desiccators w ith the crucible tongs.When the weigh
dropping to the room temperature , weigh it.To
continue to dry samples according to above-mentioned
i t is , until it is permanent.Every sample calls 0.1
g ram precisely for future spending.
2.2 Content of protein determining
According to make ni trogen of K.Covert into
the content of protein[ 7~ 9] .
2.2.1 Digestion Choose four 50ml toper bot tle , 1
and 2 add samples , 3 and 4 add distilled w ater make
contrast.To add dense sulphuric acid and cataly st
meanwhile , then digest on the electric stove in the
fume hood , until the digestive juice gets light green ,
To cont inue heating fo r 30 minutes again , washing
the bot tle wi th the distilled w ater , get the solution to
the bot tle , To get the liquid to the scale , Mixing it ,
reserving .
2.2.2 Distillation Fetch several toper bott les of
50ml , add 5ml boric acid and indicato r , The liquid
is purple.To connect it to distillation device prepare
the distillation solution.Put 10 ml of 30%NaOH into
ref lect ing room by that measuring cylinder.Heating
the steam generator begins to distillate , When the
liquid of the toper bot tle become green from purple ,
Timing for 10 minutes of dist illation from changing
color , To get the condensation in the peak to propose
the liquid , let the steam continue w ashing for about 1
minute , With w ater washing peak t one minute later.
Then carry on the tit ration w ith the hydrochloric
acid.It is the micrometric terminal point that the
indicato r solution of bo ric acid is turned bluish-greenly
into the purple.The amount of to tal nitrogen
calculated , convert to the protein amount again.
2.3 Survey of amino acid
Adopted Hitachi amino acid automatic analysis
inst rument.Accurate to claim and fetch dry to
permanent in w eight sample dry powder ration , Add
6NHCL solut ion , Hydrolysis for 22 ~ 24 hour in
110℃filters.Filt rate at low temperature steam and
do , dissolve and among w ater of a certain amount ,
dilute to certain density , reserve.It is 50ul to enter
the machine.
3 Results
Different to gather issues of seed pro tein content
of Acer truncatum Bunge determine result meet

4 Discussion
(1) Through analy sis YBF at the different
g athering protein content , results of issues of seed
indicate that f rui t seed in October content of protein
is 47.95%, secondly i t is 40.96% in December
behind fallen leaves , protein of f ruit is 35.09%.To
make use of the protein , the best gather is in
October.
(2)Through analy sis the same generic plants
YBF different seed of kind content of protein ,
Content of the SJF and SRQ is far higher than other
kinds.So w e think while developing and utilize the
seed protein of YBF , not be ignored w ith belonging
to the three plants.
(3)Through analysis YBF seed of different
g ather issues of amino acid content , three-gathering
total content of amino acid have no difference
basically to ingot.Make up of amino acid included ,
have no dif ference basically.The three time are high
in the Glu.and Arg., and contain 7 kinds necessary
amino of the human body namely Lys.Val.Met.
Ileu.Leu.Thr.Phe.[ 10] .Half the human body
must there are two kinds of amino acid in seed namely
Arg.and His.
(4)Through analy sis the same generic plants of
YBF content of the amino acid analy sis.The to tal
content of amino and acid in SRQ and CHYQ and
SJF is g reater than 30%, and is higher than YBF.
Make up from total amino acid , the Glu.and Arg are
higher than the other amino acid.Content analy sis
the human body essential of amino acid , SRQ of Thr.
Val.Leu.is too higher than other kinds .The
content of Mer.Phe.Lys.in CHYQ are higher than
in other kinds.The content of Ileu.in SJF is higher
than o ther plants.
(5)As a whole , the seed of YBF and the same
plants contain the human body necessary 7 amino
acid.And the Glu.is obviously higher than o ther
amino acid , so , we think while to develop and utilize
the YBF .It can no t igno re either of the same generic
plants.

天然油菜素内酯对侧柏种子发芽及
下胚轴伸长的影响

摘 要:采用种子发芽力和下胚轴伸长测定的方法 ,研究了天然油菜素内酯对侧柏种子发芽及下胚
轴伸长的影响.结果表明 ,不同浓度的天然油菜素内酯处理对侧柏种子的发芽率 、发芽势和平均发
芽速度均有明显的促进作用.与对照相比 ,0.4 mg/L 浓度的天然油菜素内酯处理使发芽率提高了
21.5%,发芽势提高了 13.33%,发芽时间缩短了 1.91 d.天然油菜素内酯处理也促进了侧柏种子
下胚轴的生长 ,处理 5d内 ,下胚轴伸长生长比对照高出 8.28%~ 147.64%.

Effect of natural brassinolide on seed germination
and Hypocotyl elongation of Platycladus orientalis

Abstract:The germination capacity of Platycladus oriental is seeds is obviously improved af ter the
seeds are soaked w ith the dif ferent concentrations of natural brassinolide (NBR).Compared w ith
control , the germination rate and germination energy of the seeds are increased 21.5% and 13.33%
respectively , the germination time of the seeds is shortened by 1.91 day , after the seeds are t reated
using 0.4 mg/L concentration of natural brassinolide.The hypocoty ls of Platycladus orientalis are
obviously increased post to treatment wi th the natural brassinolide.And compared wi th control , the
leng ths of Platycladus orientalis hypocotyls are increased by 8.28%~ 147.64% f rom the f irst day to
the fifth day af ter t reatment.

油菜素内酯类(BRs)是目前国际上最新型的高
效植物生长调节剂 ,可分为人工合成产品和天然产
品.人工合成产品系根据油菜素内酯的化学结构合
成 ,主要有油菜素内酯 、表油菜素内酯 、高油菜素内
酯和表高油菜素内酯等;天然产品是用萃取法直接
从油菜等 30多种植物中提取的 ,其生物活性是人工
合成产品的 5倍左右.有关油菜素内酯的生理效应
及其在农业上的应用研究曾有不少报道[ 1~ 5] .周爱
清研究了表油菜素内酯对水稻种子发芽的影响[ 6] ,
徐如娟等探讨了表油菜素内酯对黄瓜下胚轴伸长的
影响[ 7] .但在林业上的研究报道甚少.我们以侧柏
(Platycladus orientalis)种子为材料 ,研究了天然油
菜素内酯(NBR)对其种子萌发及下胚轴伸长的影
响 ,以期为播种育苗 、直播造林或飞播造林时种子处
