Abstract:Ku protein participates in DNA damage repair and responses to irradiation in both animal and plant cells. Its function and expression closely related to the ability of DSBs repair of cells. To study the Ku protein in wheat, we cloned the complete cDNA sequence of wheat Ku70 and Ku80 ,named TaKu70 and TaKu80 , respectively. TaKu70 /TaKu80 codes wheat Ku70/Ku80 protein, with 626 and 706 amino acid residues, respectively. Multiple sequence analysis showed that the protein amino acid sequences that were coded by wheat TaKu70 have 94% and 78% homology with Ku70 protein in rice and Arabidopsis respectively; The protein amino acid sequences that were coded by wheat TaKu80 had 85% and 69% homology with Ku80 protein in rice and Arabidopsis respectively. Domain and functional analysis showed that the wheat Ku protein contained the core component - Ku70 / Ku80-core domain, and the vWA domain ,which was only exist in the eukaryotic cell.
