Genetic analysis of diamondback moth, Plutella xylostella, with different susceptibilities to deltamethrin by cDNA representational difference analysis
Abstract:Differentially expressed sequences between Plutella xylostella susceptible strain and the deltamethrin-resistance strain were studied. The improved cDNA representational difference analysis was performed using cDNA from the P.xylostella deltamethrin-resistance strain as Driver amplicon and cDNA from the susceptible strain as Tester amplicon. After four rounds of subtractive hybridization, one different product with a size of about 200 bp was obtained. The fragment was cloned with pMD18-T vector and 10 clones that picked up by random were sequenced. Comparisons between our sequencing result and data in GenBank showed that there was one sequence appearing relatively high homology to ubiquitin gene, but we cant conform that it was correlated to insecticide resistance at present.
