Abstract:Pululanaseisakindofdirectendoactingdebranchingenzymethatspecificalycatalyzesthe
hydrolysisofα1,6glucosidiclinkagesofunmodifiedsubstratetogivelinearoligosaccharides,andmainly
usedinfoodindustry.However,sofartheavailablepululanaseisyetlimitedtomeettheindustrial
requirements.Resentdevelopmentsinadvancedbiotechnologyhaveprovidedsomeapproachesforhighly
eficientscreening,expressionandengineeringofpuluanasewithindustrialproperties.Otherwise,the
substratemodificationandtheimmobilizationareadoptedtoimprovethecatalyticfunctionoftheenzyme
atsomelevel.Thispapermainlysummarizestheresearchadvancesindiscovery,expressionand
engineeringofpululanaseforthedevelopmentofitsapplicationpotential.
