Abstract:Trehalose-6-phosphate synthase gene (trehalose-6-phosphate synthase, TPS) is the key gene in trehalose synthesis pathway, which plays an important role in the freezing tolerance of plant. To analyze the differences in the expression of TPS between wheat cultivars and determine the key effect of TPS gene on cold tolerance of wheat, expression differences of 7 TPS genes under a series of low temperature treatments in qRT-PCR were analyzed between cod hardy variety Dongnongdongmai1(D1) and cold-sensitive variety Chinese Spring (CS) in this experiment. The results showed that TPS4 and TPS7 were higher expressed in D1 at the beginning of the cold acclimation. The expression of TPS1, TPS2, TPS4, TPS6 and TPS7 in D1 was significantly higher than that in CS in the late period of cold acclimation. TPS2, TPS3 TPS5 and TPS 6 were reached the highest relative expression in the middle and later periods of the cold acclimation. The relative expression of TPS1, TPS6, TPS7 were higher in D1 than in CS under the treatment of-16℃, which were 72.8, 308.68, 32.83 times, respectively. In D1, the expression ofTPS3 was down regulated in 6 of 7 cold treatments, and TPS7 was up-regulation in all treatments. The result of the study had improtant significance to the breeding of new varieties of high cold resistanc.
