摘 要 ：为了更好地确定不同处理方法对样品稳定同位素分析的影响,以斑头雁(Anser indicus)为研究对象,对比了不同酸化条件、干燥方法和粉碎形式对组织材料稳定同位素碳、氮比值的影响。结果表明,不同温度和盐酸浓度对卵壳δ¹³C值有影响,80℃较40℃、60℃条件下得到的卵壳δ¹³C值富集,3 mol·L^-1 HCl较1、5 mol·L^-1 HCl浓度条件下得到的卵壳δ¹³C值贫化;冷冻干燥和烘干2种干燥方法对肌肉稳定同位素δ¹³C、δ¹⁵N的影响均未达到显著水平,液氮研磨和直接剪碎2种粉碎形式下测得的羽毛δ¹³C、δ¹⁵N值差异不显著。本研究为使用稳定同位素技术开展鸟类学相关研究提供了参考依据。

Abstract:In order to better understand the effect of different processing methods on stable isotope analysis, this research used Bar-headed Goose (Anser indicus) as an object, compared the influence of stable carbon and nitrogen isotopes ratios by different acidic conditions, drying and crushing methods. The results showed that different temperature and concentrations of hydrochloric acid affect δ¹³C ratio in egg shell. The δ¹³C in egg shell under the condition of 80 ℃ was more positive than that under 40 ℃ and 60 ℃. HCl solution of 3 mol·L^-1 was more negative than HCl of 1 mol·L^-1 and 5 mol·L^-1. The two methods of freeze drying and oven drying had not reached significant level on δ¹³C and δ¹⁵N of muscle tissue, liquid nitrogen grinding and directly cutting up had no significant effect on δ¹³C and δ¹⁵N of feathers. The results provide a reference basis for stable isotope technique applied in the research of Ornithology.
