提要: 本文着重概述了不同植物中花青素合成WD40类转录调控因子的研究进展。

1 引言
花青素是植物次生代谢产物, 属类黄酮化合
物。花青素的合成对于植物来说非常重要, 花青素
不仅是保护性物质, 有助于植物应对环境胁迫, 如
紫外线照射、营养元素缺乏以及低温损伤等
(Winkel-Shirley 2001; Teng等 2005; Cominelli等
2008; Lillo等2008; Olsen等2009; Rowan等2009),
而且, 花青素也能从分子水平上调控植物的次生生
长(Broun 2005), 其积累往往标志着某些发育阶段
的开始(Schneitz等 1995; Debeaujon等 2003)。花
青素的合成途径在基因水平上已经研究得非常清楚
(Forkmann和Martens 2001)。花青素合成的相关
基因不仅随着植物发育受到调控, 而且还在各种生
理和非生理胁迫下, 受到各种转录调控因子的复合
调控。各种转录因子所形成的复合物可以调控黄
酮类色素在特定组织中的合成, 从而调控结构基因
的表达, 以应对各种刺激(Gonzalez等 2008)。
自从由玉米(Zea mays)中克隆到第1个转录因
子 C1 (colorless 1, Cone等 1986), 人们就开始了对
花青素合成的转录调控因子的研究。起初人们认
为在植物中花青素转录激活基因由Myb和Myc两
类组成, 二者的存在是开启花青素结构基因时空表
达的关键。但1997年de Vetten等在矮牵牛(Petunia
hybride)中发现了AN11 (anthocyanin 11), 随后在
1999年Walker等发现了TTG1等WD40家族调控
因子。WD40转录调控因子的发现使得花青素调
控机制的理论研究得到进一步的完善。目前, 已有
多种花青素合成相关WD40重复蛋白得到鉴定。
2 WD40蛋白的结构特征
WD40蛋白是一类大的蛋白家族, 这类蛋白结
构高度保守, 一般含有 4~16个串联重复的WD基
元。 WD基元存在于真核生物的1%~2%蛋白质中
(Madrona和Wilson 2004), 是一个高度保守的核心
区域, 每个WD基元含有大约由40个氨基酸残基组
成的保守序列, 该序列以N末端11~24个残基处GH
二肽(Gly-His, GH)开始, C末端以WD结尾(Trp-
Asp, WD) (Neer等 1994; Smith等 1999)。除了序
列的相似性, WD40蛋白表面氨基酸残基序列亦显
示高度相似性, 并且在折叠以及空间结构等很多方
面均相当保守(Yu等 2000)。最典型的WD40蛋白
是Gβ亚基。Gβ亚基呈 7片扇叶式螺旋桨结构, 包

含 7个WD40基元(Ramsay和Glover 2005), 每个
基元的 4 个反式折叠组成 1 个片层( S onde k 等
1996)。Chothia等(1997)发现, 至少 4个重复的WD
基元才可以形成 1个完整的 β螺旋桨结构, 它通过
1或 2个片层参与WD40蛋白和其他蛋白的互作
(Haar等 1998)。该结构域赋予WD40蛋白家族成
员一个共同的特征: 介导蛋白质之间的互作、调整
多蛋白复合物的装配, 其重复的WD基元作为蛋白
质互作的支架, 可以同时参与几个蛋白质的相互作
用(Smith等 1999; Tyers和Willems 1999), 但是缺
乏DNA结合区域, 并且没有任何酶类的催化功能
(Ramsay和Glover 2005)。
目前, 有关植物WD40蛋白的研究还不多, 主
要局限于拟南芥(Arabidopsis thaliana) WD40蛋白
家族。序列预测拟南芥基因组有 269个WD40基
因, 可分成 143个亚家族(van Nocker和 Ludwig
2003), 其中的 113个亚家族成员在酵母、果蝇和
人类中都存在同源基因(Skurat和Dietrich 2004), 它
们在花青素合成、分生组织形成、幼苗发育、花
发育、光信号传递和感知等方面具有重要的作用
(Ullah等2003; Guitton等2004; Yamagishi等2005)。
除拟南芥外, 已经从矮牵牛、玉米、紫苏(Perilla
frutescens)、紫罗兰(Matthiola incana)等中克隆到
部分编码WD40蛋白的基因。下面介绍这些基因
在花青素合成中的功能作用。
3 花青素合成中的WD40蛋白
3.1 矮牵牛AN11蛋白 AN11含有5个WD重复基
元(Neer等1994), 每个重复基元中包含保守的氨基
和羧基终端的核心结构, 这些核心结构被短的氨基
酸序列分隔开来。
细胞碎片实验显示AN11蛋白定位于细胞质
中, 推测其对转录过程没有直接调控作用。在矮牵
牛 an11突变体中, 结构基因DFR (dihydroflavonol-
4-reductase)的表达量下降, 但只影响花的色素积累,
组成型表达 AN2可以恢复 an11, 恢复DFR的表达
活性, 这说明在花青素合成的调控途径中 AN11位
于 AN2的上游(de Vetten 等 1997)。进一步研究证
明AN11的转录与MYB类转录因子AN1和AN2的
转录之间没有关系(de Vetten等 1997; Quattrocchio
等 1998), 然而, de Vetten等(1997)推测AN11有调
控MYB类调控蛋白的活性作用。AN11在无色素
形成的植物组织中也有表达, 并且在植物和动物的
进化的过程中都表现出高度的保守性, 因此在调控
花青素的合成外, AN11可能还具有更为广泛的功
能(Joseph等 1998), 如调控种子表皮细胞的形态等
(de Vetten等 1997)。
3.2 拟南芥TTG1 (transparent testa glabra1)蛋白
TTG1与矮牵牛AN11具有高度的同源性(Springob
等 2003)。拟南芥 TTG1可以调控类黄酮 /花青素
的合成(Ramsay和Glover 2005)、种皮的发育以及
叶表皮毛的形成(Larkin等 1994, 1999; Walker等
1999; Payne等 2000; Ramsay和 Glover 2005)。
TTG1基因突变严重地影响所有这些发育过程, 表
明了 TTG1的广泛调控功能(Broun 2005)。TTG1
影响 DFR的功能, ttg1突变体的 DFR催化受阻,
TTG1可以诱导DFR的表达(Heller等 1985)。玉米
的R基因编码一个 bHLH (basic helix-loop-helix)蛋
白 TT8 (transparent testa8), 与MYB转录因子互作,
控制花色素的合成, R基因能够补偿ttg1的突变, 可
能在TTG1下游行使功能(Spelt等 2000)。TTG1通过
与 bHLH类转录因子GL3 (glabra3)、EGL3 (enhancer
of glabra3)和TT8 (Zhang等2003)以及MYB类转录
因子拟南芥 TT2 ( transparent testa2)或 PAP1
(production of anthocyanin pigment1) (Baudry等
2004; Ramsay和Glover 2005)相互作用来实现控制
花青素结构基因的时空表达(Nesi等 2001)。
TTG1 C末端区域对于该蛋白功能发挥非常重
要(Walker等 1999)。Matsui和Ohme-Takagi (2009)
研究显示, TTG1 C末端氨基酸序列中第 309个至
第341个氨基酸对于它和TT8的相互作用从而调控
种皮颜色是必需的。
3.3 玉米PAC1 (pale aleurone color1)蛋白 在玉米
中约有20个基因影响玉米花色素的产生, 其中pac1
编码WD40蛋白(Ludwig等 1989; Selinger和Chan-
dler 1999; Carey等 2004), 类似于花青素调控蛋白
AN11和 TTGl (Selinger和 Chandler 1999; Carey等
2004)。pac1缺失突变体中, pac1的缺失导致 a1、
bz1和 c2这些花色素苷结构基因的表达都下调, 在

种子的糊粉层没有花青素的积累, 但是种皮的花青
素合成并不受到影响, 这点与 ttg1不同(Selinger和
Chandler 1999; Carey等2004)。可能由于基因的遗
传丰余现象等原因, PAC1在种皮花青素合成中的
作用并不明显。由于 ttg1能充分互补 pac1功能,
PAC1和 TTG1在功能上应是等价的, 因此, PAC1
和TTG1这2种调控蛋白在生物学功能上的差异可
能是由与TTG1和PAC1相互作用的不同调控因子
以及它们共同形成的相互作用导致的, 而不是由
TTG1 和 PAC 1 本身的功能差异引起的(Br oun
2005)。
in1 (intensifier 1)是花青素结构基因表达的抑
制基因, 它编码的蛋白具有 bHLH结构, in1突变会
促进谷粒中花色素增强(Burr等1996), pacl处于in1
的上位调控, 应答于玉米种子糊粉层、角质鳞片以
及根部中花青素的生物合成。
3.4 紫苏PFWD蛋白 Yamazaki等(2003)在紫苏叶
子中发现了花青素合成相关的PFWD蛋白, 在拟南
芥中过量表达 P F W D 可以增加花青素的合成。
PFWD含4个WD重复序列, 氨基酸序列与AN11和
TTG1分别有 81.3%和 77.8%的相似度, 相当保守,
在 PFWD的 3非转录区有 4个TATT基序, 增加了
mRNA的稳定性(Reeves等 1987)。PFWD蛋白存
在许多翻译后修饰, 最典型的是N-糖基化、磷酸
化以及烷基化。真核生物中, N-烷基化蛋白主要
铆定于原生质膜或者其他细胞内膜上(Resh 1999)。
PFWD蛋白可能与膜系统相关。PFWD N末端的
一段氨基酸序列与核定位信号NLS序列相似, 但亚
细胞定位实验证明, 该蛋白定位于细胞质中(Stacey
等1999), 推测PFWD可能通过与MYC家族蛋白共
同作用, 可从细胞质中转移到细胞核上, 在花青素
合成等信号转导途径在起着信号传递的作用
(Yamazaki等 2003)。
PFWD在各种组织中广泛表达, 即使是无色素
积累的组织中, PFWD cDNA序列在红色和绿色紫
苏中无核苷酸差异。PFWD可能与拟南芥 TTG1
相似, 参与多重调控途径。拟南芥中过量表达
PFWD明显增多了表皮毛的数量, 但TTG1过量表
达却对表皮毛数量无影响(Stacey等 1999)。同时,
PFWD的过量还影响到植物的其他发育甚至对植物
是致死性的伤害, 相比之下, 其他的转录因子, 如
MYB或者MYC家族成员的作用途径较窄, 只作用
于花青素的积累。PFWD蛋白可能负调控发育过
程中一些重要蛋白(Yamazaki等 2003)。
3.5 紫罗兰TTG1蛋白 早在1949年, Kappert就已
经发现紫罗兰 e、f、g位点参与花青素的合成。
其中 e位点编码一种典型的WD40重复蛋白, 该基
因 3非翻译区没有内含子, 与拟南芥 ttg1编码区核
苷酸序列有 85.3%的相似性, 所编码的蛋白质与
TTG1 蛋白有高达 96.2% 的同源性(Dressel和
Hemleben 2009)。
紫罗兰 e位点突变株系 17 (W158R)只有一个
核苷酸发生突变, 致使在WD模块中的色氨酸突变
为精氨酸, 导致该蛋白的调控功能缺失, DFR不表
达, 花青素的合成阻断, 根毛形成以及种皮发育和
色素形成也受到阻断, 开白色花朵并且表面光滑。
3.6 棉花(Gossypium hirsutum) GhTTG蛋白 Hum-
phries等(2005)从棉花中也克隆到4个TTG1基因的
同源体GhTTG1~4, 且这 4个WD40基因与矮牵牛
AN11、紫苏PFWD等也表现了高度的相似性。而
且, GhTTG1和GhTTG3基因除了能够恢复拟南芥
ttg1突变体的毛状体的形成, 也能够弥补开白花的
ttg1突变体中花青素的缺陷, 同时它们在棉花纤维
的起始、分化中具有重要的作用, 可能替代 TTG1
在植物毛状体起始和花青素途径中作用。因此, 推
测GhTTG1和GhTTG3在拟南芥毛状体和棉花纤
维的发育过程中的作用机制类似。
3.7 油菜(Brassica napus) BnTTG蛋白 利用同源克
隆策略从油菜中克隆了2个编码WD40调控蛋白的
基因。它们分别被命名为BnTTG1-1和 BnTTG1-2,
NCBI保守域预测的结果表明 2个基因均有WD40
的保守域以及相应的基元, 其二级结构符合WD40
转录因子的结构特征。半定量 R T - P C R 表明,
BnTTG1-1和 BnTTG1-2 的表达与 AtTTG1很相似,
在油菜的主要组织器官都表达, 可能在油菜种皮色
素的形成中有重要的调控作用(Lu等 2007)。
4 WD40蛋白的进化
WD40重复蛋白是一个古老的蛋白家族, 从动

物(包括两侧对称动物和水螅)、真菌、粘菌盘基
网柄菌到植物, 在所有的已经测定的真核基因组中
都存在(van Nocker和 Ludwig 2003), 这种普遍性
说明现代真核生物WD40家族起源非常早, 但是在
原核基因组中, 只在鱼腥藻、蓝藻以及嗜热单孢菌
属有发现(http://bmerc-www.bu.edu/wdrepeat/), 而
大肠杆菌、其他细菌基因组中以及古细菌中无
WD40重复蛋白, 这意味着原核WD40蛋白可能起
源于古老的蓝藻类细菌(或者可能是线粒体起源的)
(Martin和 Russell 2002)。
在所有研究过的WD40蛋白中, WD结构域都
是作为蛋白质互作的支点, 但是这个家族蛋白的作
用范围相当广泛, 并且具有选择性, 在不同的领域
与不同的蛋白质相互作用。较之植物, 在动物基因
组中, 该家族蛋白有更多的重复单元, 增加了其功
能多样性(van Nocker和 Ludwig 2003)。但是在动
物WD40家族还没有发现可以与MYB和bHLH相
互作用的成员, 只脊椎动物c-Myc被认为具有激活
WD40的功能(Bishop等 1991)。在植物界多种高
等植物表皮表型的调控中WD40家族蛋白与MYB
和bHLH形成的这样一个调控网络调控可以追溯至
1 2 亿年前。
花青素合成中WD40蛋白都属于同一个进化
支, 并且在功能上, 它们都能通过形成特定的结构
与其他蛋白质相互作用。在拟南芥 ttg1突变体中
易位表达拟南芥TTG1蛋白的类似WD40蛋白可以
弥补突变体的很多表型, 这表明这些蛋白的作用机
理相似, 都能与某一类蛋白结合发挥作用(Steven和
Philip 2003)。遗传学分析和酵母双杂交显示, 这类
调控WD40蛋白能与bHLH和MYB类蛋白共同调
控花青素的合成(Bernhardt等 2003; Payne等 2000;
Zhang等 2003)。
事实上, 在很多植物中, TTG1不仅仅调控花
青素的合成, 例如, 拟南芥 TTG1还正调控种皮色
素合成、根毛的发育、决定表皮毛的分化、负
调控子叶下胚轴气孔细胞运动(Berger 等 1998;
Walker等1999); 矮牵牛的AN11还调控种子表皮细
胞的形态(de Vetten等 1997)。与拟南芥相比, 矮
牵牛AN11和玉米PAC1只表现出微弱的基因多效
性。这可能与长期以来对这两种植物的驯化有关,
这些蛋白失去了一些性状的控制功能, 只局限或者
集中在花青素合成的调控中, 而在拟南芥中却得到
进一步发展(Broun 2005)。
在拟南芥和紫罗兰中, 编码花青素合成酶的结
构基因, 如 CHS (chalcone synthase)、DFR中GC
含量相似, 对紫罗兰 ttg1和拟南芥 ttg1碱基GC含
量做一个比较显示, 紫罗兰 ttg1有更丰富的GC含
量, 分别为 54.1%和 46.1%, 拟南芥中较大一部分
C碱基位点被中性的T取代。这种GC含量的差异
却没有显著的影响到氨基酸组成, 并且这种高比例
的GC值不是生态型, 而是进化的结果(Torres等
1990)。此外, 单子叶植物, 玉米花青素合成WD40
家族成员 pac1的基因也有更高的GC含量。有些
TTG1蛋白在功能上还不能确定, 如苹果属和松叶
菊属。进化树对这些 TTG1以及类似 TTG1蛋白
分析显示紫罗兰TTG1、拟南芥AtAN11以及棉花
TTG2和 TTG4 (Humphries等 2005)、玉米MP1
(Carey等2004)代表的是WD40家族中的另一个分
支。
5 WD40蛋白的调控机理
WD40蛋白是高度保守的(在不合成花青素的
生物体中也如此)。目前, TTG1及类 TTG1蛋白
的作用模式还不清楚。只知道这类WD40蛋白的
表达相当广泛, 可以与bHLH类以及MYB类调控因
子广泛的作用, 定位于细胞质或细胞核中(Baudry等
2004; de Vetten等 1997; Sompornpailin等 2002)。
具体的分子功能仍不清楚, 其作用机制并不能简单
的用它们所包含的WD序列来定义。
通过酵母双杂交实验推测WD40类转录因子
可能介导与其作用的调控因子从细胞质转移到细胞
核中, 如矮牵牛AN11、紫苏 PFWD定位于细胞质
中(de Vetten等 1997), 然而当PFWD与 bHLH类调
控因子(PFMYC)共同作用时, 一部分PFWD蛋白转
移至细胞核中, 这意味着这类WD40蛋白可能直接
参与到转录水平的激活中(Zhang等 2003)。拟南
芥TTG1与MYB类转录因子(AtTT2)和bHLH蛋白
(AtTT8)共同作用可以促进转录。AtTTG1增加一
个转录激活结构域可以增强效果, 显示AtTTG1可

以参与到转录复合体中(Baudry等 2004)。然而
TTG1可能不是bHLH以及MYB类转录因子的核定
位所必需的, 因为bHLH类转录因子的过量表达可
以回复 ttg1突变体中 TTG1的功能缺失(Payne等
2000)。
拟南芥TTG1多效型调控多种生理功能, 可以
解释为TTG1作为蛋白相互作用的支架(Ramsay和
Glover 2005)。在玉米、拟南芥和矮牵牛中的研
究表明, WD40蛋白可以和MYB以及bHLH蛋白相
互作用, 以WD40/Myb/bHLH (WMb)的调控模式共
同调节花青素的合成(de Vetten等1997; Quattrocchio
等1998; Walker等1999; Spelt等2000, 2002; Morita
等 2006; Schwinn等 2006)。
在WMb调控复合体中, TTG1仅仅是作为介
导MYB和bHLH相互作用的桥梁, 还是在细胞核中
起着更为活跃的功能?一些酵母和人类WD40蛋
白, 如果蝇调控蛋白GRO (Groucho)以及它在人类
中的同源物 TLE1 (Fishe和 Caudy 1998; Chen和
Courey 2000)同TTG1一样, 与bHLH类转录因子相
互作用参与基因的调控网络。与之不同的是, GRO
和TLE1通常都与转录抑制因子一起形成负调控复
合体。然而, 有趣的是, 这 2种蛋白还可以与具有
Runt结构域蛋白作用, 而该蛋白既可以作为靶基因
的激活子也可以作为抑制子, 这取决于靶基因启动
子(Aronson等 1997)。尽管 TTG1与GRO/TLE1所
含WD40基序不同, 但是在结构预测上它们很相近:
C末端都有β-螺旋桨结构, 可以作为蛋白相互作用
的支点(Walker等 1999)。GRO的一个很重要的特
点是可以通过中心结构域中未乙酰化的组蛋白与染
色质不同位点结合形成抑制转录的环境(Chen等
1 9 9 9 )。已经发现了一些参与染色质重塑植物
WD40蛋白, 如 AtMSI1 (Arabidopsis thaliana
multicopy supressor of Ira 1), 它可以与含 6个WD
单元的 FIE (fertilization independent endosperm)以
及Medea (polycomb-group protein MEA)相互作用,
使染色质处于抑制转录的环境, 从而使得植物在受
精之前抑制形成胚乳, 调控种子的发育(Luo等
1999; Ohad等1999; Köhler等2003)。它们与TTG1
的同源性较低但极显著。同样的, TTG1可能也参
与到染色质重塑中, 克服染色质抑制转录。其作用
机制可能是与在和bHLH-MYB形成复合体中作用
相同。TTG1可以直接与染色质重组蛋白相作用,
如组蛋白乙酰基转移酶, “打开”染色质, 或者是干
扰染色质重塑系统中抑制转录的调控因子(Broun
2005)。
6 展望
目前, 虽已在矮牵牛、拟南芥、玉米、紫苏、
紫罗兰等多种植物中鉴定了花青素合成相关的
WD40类转录因子。但要全面认识WD40类转录因
子的功能还有许多工作要做, 包括以下几个方面。
(1)各物种中WD40类转录因子的鉴定。WD40
重复蛋白是一个古老的蛋白家族, 随着更多生物基
因组测序工作的完成, 将会有更多生物的WD40基
因序列得到鉴定, 这将为研究不同生物WD40类转
录因子的进化关系提供良好的数据, 也为进一步阐
明整个WD40大家族的进化历史以及研究基因结
构进化历史提供更多的分析材料。
(2)花青素合成相关的WD40类转录因子的功
能研究, 以及各转录因子之间如何相互作用并共同
调节花青素的生物合成。
(3)通过WD40类转录因子基因的遗传转化控
制花卉和果实的颜色, 实现品种改良。
