Abstract:The main objective of this study was to identify and map the Cercospora sojina race 15 resistance gene in soybean Kenfeng 16 with simple sequence repeat (SSR) markers. Soybean plants of Suinong 10 (susceptible), Kenfeng 16 (resistant) and F2 and F3 populations derived from a cross of these two cultivars were screened for the inheritance analysis of C.sojina race 15 resistance. SSR markers linked to the C.sojina race 15 resistance gene were identified using bulked segregation analysis. Furthermore, 108 soybean breeding lines were analyzed for the marker efficiency. The results showed that resistance to C.sojina race 15 in Kenfeng 16 was controlled by one pair of dominant genes, which located on linkage group J of soybean genome and was named Rcs15. The result of linkage analysis by using Mapmaker/Exp 3.0 b software showed that five SSR markers, Satt 529, Satt 431, Sat_151, Satt 547 and Sat_224, were tightly linked to the resistance gene. The order and genetic distance of the markers in the resistance gene locus were Sat_151-10.7 cM-Satt 529-18.5 cM-Rcs15- 6.7 cM-Satt 547-7.8 cM-Sat_224-10.7 cM-Satt 431. Conformance analysis showed that the screening efficiency of markers Satt 547 and Sat_224 was over 85%. These two SSR markers are useful for molecular marker assisted selection for C.sojina resistance and soybean breeding program.
