Abstract:To identify the teliospores of genus Tilletia intercepted from stem debris of wheat, both the nested PCR and real time PCR specific to Tilletia indica were used with the sequence analysis of the PCR products from universal and specific primers. The result showed that the teliospores in stem debris of wheat got positive reaction with T.indica-spicific primer I9/I4 in nested PCR and T.indica-spicific probe TIND in real time PCR, respectively. The sequence analysis of PCR products revealed that the sequences of ITS region and PCR product from T.indica-spicific primer I9/I4 had 100% identity with those of T.indica. Based on the morphological characteristics, nested PCR detection, real time PCR assay and sequence analysis of PCR products, the teliospores from stem debris of wheat are identified as Tilletia indica.
