Abstract:In order to provide technical support for breeding new varieties and germplasm innovation of Chinese cabbage resistance to black spot, the black spot leaves samples of Chinese cabbage were collected in Shaanxi Province. The pathogen was isolated and purified by single spore and identified by special molecule marker. It was determined that Alternaria brassicicola was the only pathogen causing black spot in spring-summer Chinese cabbage. By studying the influence of inoculation spore concentrations, seedling stage and growing temperature on black spot occurrence with three different resistance varieties, an artificial inoculation method for black spot identification was optimized and set up, which key procedure is praying 1×104pfu/mL spores on seedling with two true leaves, and placing at 25℃ condition, then keeping moisture and dark for 24h, keeping moisture at night from 4th-7th day, keeping moisture 24h in 8th day, then investigated. The result from the method was exact in accordance with the true resistance type of Chinese cabbage.
