Abstract:The ovaries in vitro of Yucca gloriosa were used as the explant inocalate material in order to induce ovaries to expand and callus to form in MS medium supplemented with KT, 2,4-D, and LH. A large number of thickly growing buds were sprouted in MS medium supplemented with 6-BA, NAA and LH, and then transplanted to MS medium supplemented with IBA so as to induce rootlets to form. The transplant survival rate reached more than 72% in soil medium sterilized by intermittent high temperature and mixed with germicide. A lot of complete plantlets had been obtained. Observing under microscope, the chromosome number of test tube plantlets is the same with that of original root sprouts. The peroxidase activity of every differentiation periods in culture is determined. The results show that the activity has a positive relationship with differentiation progress.
