生物医学新兴学科与中药现代化 现代组合成分药物的研究

摘要:中药的特色是通过多成分、多靶点的整体调控作用来系统纠正疾病造成的机体失衡。中药在治疗多基因、
多因素引起的某些复杂疾病方面相对西药(通常是单一化学成分药物)有独到优势。但以天然动植物为来源的中药
在质量控制和疗效科学依据方面的不足严重影响了中药在现代社会的应用。目前,组合治疗现代医学和系统生物
学的发展为中药现代化提供了新的机会。提出应用现代组合成分药物和组合治疗新概念,科学阐明中药药效的学
术观点;并提出综合应用蛋白质组学和化学生物学等生物医学新兴科技从中药中开发现代组合成分新药的技术。

Abstract:Chinesemateriamedica(CMM)defineshealthasthebalanceofwholebodysystemand
medicationsarede ignedtOrestorethisbalance.UnlikewesternmedicinethatgenerallyUsesasingle
chemicalentityandsetsasinglephysiologicaltarget,theefficacyherbalformulationsinCMMresultsfrom
complexmixturesofnumerouschemicalcomponentswhichsimultaneouslyinteractwi hmultiplemolecular
targets.CMMclinicallyshowstheadvantagesoverwesternmedicineinthetreatmentofsomemulti—gene
andmuhifaetordiseases.yettheadoptionofCMMstemmedfromnaturalanimalsndplantsinmodernso—
cietyisimpededbylackofqualitycontrolandabsenceofscientificprooftheireffectiveness.Currently.
thedevelopmentofcombinatorialmed cinesa dsystembiologyprovidenewopportunitiestomodernizethe
CMM.Thus,thisreviewfocusesonthemodernizationofCMMbyemployingtheconceptofcombination
therapyndcutting—edgebiomedicaltechnologiessuchasproteomics,bioehip,andchemicalbio ogy.
Moreover,theau orputsforwardthenewideasndmethodologiesforthedi coveryofmodernmulticom—
ponentdrugsfromCMM. .

中药是中华民族几千年文明的宝贵遗产。从已
经千百年临床验证的中药中开发自主知识产权的新
药,是利用我国独有的中医药宝库进行现代新药创
制的捷径,全新抗疟特效药青蒿素及其衍生物的研
发成功就是典型范例[1]。建国以来,我国科学家利用
提取和结构分析技术对常用中草药进行了系统化学
成分研究,明确了中药药效的物质基础是中药所含
的化学成分,并从中发现了以青蒿素为代表的一系
列单体化学成分新药[2]。但研究人员在研究过程中
也困惑地发现:一些中药经过提取分离和逐步纯化
后根本找不到对应中药药效的有效单体成分:在利
用生物活性跟踪中药化学成分的分离纯化时,常发
现中药成分越纯、活性越低的反常现象,甚至在纯化
过程中活性突然丢失。临床上也发现某些提纯的中
