Abstract:Wheat line Sw92 is highly resistant to Puccinia triticina in China either at seedling stage or adult plant stage. F1, F2 and BC1 generations from the crossing of Mingxian 169×Sw92 was inoculated with the dominant race PHT of Puccinia triticina. The gene for leaf rust resistance of Sw92 was detected by the genetic analysis. The results indicated that resistance of Sw92 to the race PHT of Puccinia triticina was controlled by one pair of recessive genes. The bulked segregant analysis (BSA) of F2 generations and simple sequence repeats (SSR) techniques were used to develop molecular markers linked to the resistance gene in Sw92. The resistant and susceptible DNA pools were prepared separately from the F2 plants crossing between Mingxian169 and Sw92. Two SSR markers namely WMC494 and WMC737 on 6BS of wheat chromosome closely linked to the leaf rust resistance gene was obtained. The results of linkage analysis showed that the gene for leaf rust resistance in Sw92 was located on 6BS with the genetic distance of 3.4cM to WMC494 and 15.0cM to WMC737, respectively, which probably is a new leaf rust resistance gene LrSw92(temporary designation)other than the known Lr36 and Lr53 on 6BS of wheat.
