苦荞醇提物的镇静催眠作用研究(摘要)(英文)

Abstract [ Objective] TheaimwastostudythesedativeandhypnoticefectsofTartaryBuckwheatethanolextractandlayafoundationforfur-
therexpandingtheapplicationofTartaryBuckwheat.[Method] TheefectsofTartaryBuckwheatethanolextractonpentobarbitalsodiumandmicespontaneousactiontimeswererecorded.[Result] EthanolextractofTartaryBuckwheatcouldprolongmicesleepdurationcausedbypen-
tobarbitalsodium(suprathresholddosage), increasemicesleepnumbercausedbypentobarbitalsodium(subthresholddosage), andobviously
inhibitmicespontaneousactiontimes.[ Conclusion] EthanolextractofTartaryBuckwheathasafunctionofsedation.

  TheTartaryBuckwheatisdicotyledoninpolygonaceae
anditisuniqueresourcecropforedibleandmedicinaluse.
ThemedicinaluseofTartaryBuckwheathavebeenrecorded
inChineseMedicalBooks, inrecentyears, thegrain, root,
stem, leavesandflowerofTartaryBuckwheathavefla-
vonoids, whichhavemanyvariousactivitiessuchashypogly-
cemicefect, reducingbloodlipid, antioxidation, enhancing
immunity, scavengingfreeradicals, besides, theyalsohave
therapeuticeffectsondiabetesmelitus, hypertension, hyper-
lipidemiaandcoronaryheartdisease.However, thesedation
andhypnosisofethanolextractofTartaryBuckwheathavenot
beenreportedyet, whilethesedationandhypnosisofethanol
extractofTartaryBuckwheatarestudiedinthisexperimentto
layfoundationforexpandingclinicalapplicationofTartary
Buckwheat.
MaterialsandMethods
Materials
Drugs SomefulTartaryBuckwheatgrainswithhul(colect-
edfromLiangshanYiAutonomousPrefecture, SichuanProv-
ince)wereselectedforexperiment.Theyweregroundafter
dry, andthentheywererefluxedat85℃ by75% ethanoland
extractedtwicefor2 heverytime.Thefiltratesoftwicewere
combined, thentheethanolwasrecycledandthefiltrates
wereconcentratedtopasteshape.Thepastesweredriedat
80 ℃ toobtainextractforfutureuseandtheyieldratewas
6.67%.BunaoanshenSliceswereboughtfromWuhanJian-
minPharmaceuticalGroupCORP, LTD.Aldrugswerepre-
paredby0.5%CMC-Na.
Animals TheheathmaleKunmingmicewereweightedfrom
18 to22 gandthecertificationnumberwasSCXK(Chuan)
2008-14.
Reagents  The pentobarbitalsodium wasboughtfrom
ShanghaiChemicalReagentFactory
Instrument ZZ-6 typemouselocomotoractivitywasbought
fromChengduTechnology&MarketCo., LTD.
Methods
InfluencesofethanolextractofTartaryBuckwheaton
hypnotictimeofpentobarbitalsodiumtomice 40 male
miceweredividedintofourgroupsrandomlyandevenly,
namely, controlgroup, high-dosegroupofTartaryBuckwheat
ethanolextract, low-dosegroupofTartaryBuckwheatethanol
extractandBunaoanshenslicegroup.8 hbeforeexperiment,
themicewereonlywateredbutnotfed, duringexperiment,
themiceincontrolgroupwerefedisovolumetric0.5% CMC-
Na, whilemiceinlow-dosegroupofTartaryBuckwheatetha-
nolextractandhigh-dosegroupofTartaryBuckwheatethanol
extractwerefedwith0.13 and0.5 g/kgethanolextractof
TartaryBuckwheat, however, themiceinBunaoanshenslice
groupwerefedwith2.28 g/kgBunaoanshenslice.Almice
werefedwithdrugsforsuccessive7days;however, afterfed
drugsfor30min, themicewereinjected45 mg/kgpentobar-
bitalsodium(decidedbypreliminarytest).Thesleeptimeand
sleepdisappearancetimeweretaken, thenthesleeplatency
(sleepappearancetime-recordstartingtime)andsleeptime
andsleeptime(sleepdisappearancetime)werecalculated.
InfluencesofethanolextractofTartaryBuckwheaton
hypnoticdosageofmicecausedbysubthresholddosage
ofpentobarbitalsodium Thegroupingandmedicationad-
ministrationwereasthesameastheprevious.Afteradminis-
tratingdrugsfor30 min, micewereinjected35 mg/kgpento-
barbitalsodium(subthresholddosage), thenthesleepymice
wererecorded(afterinjectedfor15 min, themicewithout
rightingreflexwasrecordedassleep.)andthesleepingrate
ineverygroupwascalculated.
InfluencesofethanolextractofTartaryBuckwheaton
micelocomotoractivities[ 2]  Thegroupingandmedication
administrationwereasthesameastheprevious.Afteradmin-
istratingdrugsfor30min, micewereputintolocomotoractivi-
tydeterminatortofor5 minadaptations, thenthelocomotor
activityin5 minwererecorded.
Datastatisticalmethod Thedatawereconductedinter-
groupttestandχ2 test.
ResultsandAnalysis

Discussions
TheTartaryBuckwheatisusedasfoodnormalyandit
hashighediblevalue.Theliteraturesinpastdynastiesalso
paymuchatentiontoediblerecord;however, themedicalre-
cordhasnotreferredinsomniatreatment.Itisfoundthatthe
TartaryBuckwheatmainlycontainsflavonoidsthroughmodern
drugresearchmeans, soitispredictedthatTartaryBuck-
wheathassedativeeffectthroughcombiningpreviousstudies
ofanalgesictest.Theexperimenthasdemonstratedthatthe
ethanolextractofTartaryBuckwheatcanprolongsleeptimeof
micebelowthresholdconcentrationandincreasethesleepy
mice;besides, italsocandeclinelocomotoractivitiesof
mice, sotheTartaryBuckwheathassedativeandhypnotic
efect.

苦荞醇提物的镇静催眠作用研究(摘要)

[目的]研究苦荞醇提物的镇静催眠作用 ,为进一步扩大苦荞临床应用奠定基础。[方法]将颗粒饱满的苦荞带壳种子干燥后适度研碎 , 75%乙醇(固液比 1∶10), 85℃回流提取 ,提取 2次 ,每次 2h,合并 2次滤液 ,回收乙醇并浓缩至稠膏状 ,经 80℃烘干 ,得苦荞提取物 ,提取物得率为 6.67%。记录苦荞醇提物对戊巴比妥钠引起的小鼠睡眠持续时间 、入睡率及小鼠
自主活动次数。将 40只雄性小鼠随机分为 4组 ,每组 10只 ,分别为空白对照组、苦荞醇提物高剂量组 、苦荞醇提物低剂量组和阳性药物对照组;试验前 8h禁食不禁水 ,试验时空白对照组给予等体积的 0.5% CMC-Na,苦荞低剂量组、高剂量组分别给 0.13g/kg、0.5g/kg苦荞醇提物 ,阳性药物对照组给 2.28g/kg的补脑安神片;连续给药 7d。①末次给药 30min后 ,小鼠腹腔注射戊巴比妥钠 45mg/kg(由预试验确定),记录睡眠出现和消失的时间 ,计算睡眠潜伏期(睡眠出现时间 -记录开始时间),睡眠持续时间(睡眠消失时间-睡眠出现时间);②末次给药
后 30min,小鼠腹腔注射戊巴比妥钠 35mg/kg(戊巴比妥钠阈下催眠量 ,即 90%~ 100%小鼠不入睡的戊巴比妥钠最大剂量 ,由预试验确定),观察并记录入睡动物数(注射后 15min内小鼠翻正反射消失达 1min以上者记为入睡),计算各组入睡率;③末次给药 30min后将小鼠放入小鼠自主活动测定仪中 ,让其适应 5min,然后开始测定 5min内自主活动次数。[结果]苦荞醇提物能延长戊巴比妥钠阈上剂量引起的小鼠睡眠持续时间 ,增加戊巴比妥钠阈下剂量引起的小鼠睡眠只数 ,且能明显减少小
鼠自主活动次数。[结论]苦荞醇提物具有镇静作用。关键词 苦荞醇提物;镇静;催眠;小鼠
