Abstract:TheoptimumtechnologyofSevagmethodwasstudiedtoremoveproteinfromPhelinusigniari
uscrudepolysaccharides,andtestantitumoractivityofpolysaccharidesbyMTTmethod.Onthebasisof
singlefactorexperiment,includingtheamountofSevagreagent,thevolumeratioofchloroformandbuta
nol,extractiontimesandshakingtime,anorthogonalexperimentwasdesigned.Afteranalysisofvari
ances,theoptimumtechnologyofSevagmethodwasdetermined.Then,antitumorexperimentinvitro
wastested.TheresultsshowedthattheoptimumtechnologyconditionsofSevagmethodwereasfolows
theamountofSevagwas50mL,thevolumeratioofchloroformtobutanolwas3,theextractiontimeswas
6,andtheshakingtimewas20min.Theresultofcelexperimentinvitroshowedthattheinhibitionto
wardcancercelEC109andSiHabetweenthecrudeanddeproteinizativepolysaccharideshadnosignifi
cantdiference.
