[Abstract] Objective:Toinvestigatetheefectsofresveratrol(Res)onneonatalratcardiomyocytelesioninducedbyhypoxia.
Method:Thecardiomyocyteofneonatalratswereculturedinvitroandthemodelofcardiomyocytehypoxiawasestablished.Thecar
diomyocytevitalitiesweredeterminedbyMTTassay,theHIF1αexpressionlevelsinmyocardialcelswasdetectedbyimmunohisto
chemical,theactivitiesofperoxidase(GSHPx)andlactatedehydrogenase(LDH)weremeasuredaswel.Result:Aftertheadminis
trationofhypoxiafor24hours,theHIF1αexpressioninmyocardialcelswassignificantlyincreased.TheLDHlevelintheculture
mediumwasincreasedfrom(93.07±15.84)U·L-1to(750.77±181.51)U·L-1(P<0.01).TheintracelularGSHPxactivity
wasdecreasedfrom(46.96±8.36)U·mL-1to(27.13±4.76)U·mL-1(P<0.05).Res25,50and75μmol·L-1coulddose
dependentlyinhibittheraisingoftheHIF1αexpressioninmyocardialcelsinducedbyhypoxia.TheLDHactivitiesweredecreased
dosedependentlyto(486.17±69.97),(189.43±32.07),(155.34±29.57)U·L-1,respectively(P<0.05orP<0.01).
TheGSHPxactivitieswereincreaseddosedependently(33.55±6.34),(37.67±6.73),(41.44±7.91)U·mL-1(P<0.05or
P<0.01).Conclusion:Reshasaprotectiveefectonneonatalratcardiomyocytelesioninducedbyhypoxia.
