摘 要 :目的:研究二氢杨梅素(DMY)对兔复发性口腔黏膜溃疡的作用。方法:取新西兰兔,背部脱毛,采用同种异体口腔黏膜匀浆与弗氏完全佐剂作成免疫抗原,脊柱两侧多点皮内免疫注射,2周1次,共5次;第5次免疫注射后7d,口腔黏膜表面给DMY 4 d;给药前和给药期间观察口腔溃疡发生次数,末次给药后次日,比浊法测血清抗口腔黏膜抗体滴度,ELISA法测血清IgG含量,取口腔黏膜做组织病理学检查并根据病变程度进行分级评分。结果:于第3次免疫注射后,兔口腔黏膜均出现溃疡,给药前溃疡发生次数为11.8±1.4(n=40);对照兔血清抗体滴度、IgG升高,溃疡部位黏膜角化层脱落、上皮细胞层和固有层大量炎性细胞浸润,病变程度计分为2.06±0.85。给DMY37.5,75,150 mg.kg-1后,口腔黏膜溃疡发生次数明显减少,溃疡部位病变减轻,病变程度计分降低(1.81±0.91,1.37±0.80,1.12±0.61);血清抗体滴度、IgG降低。结论:DMY有抗兔复发性口腔黏膜溃疡的作用。
Abstract:Objective: To study the effects of dihydromyricetin on recurrent oral ulcers of rabbits.Method: Forty-eight of New Zealand rabbits were randomLy divided into 6 groups.The model of oral ulcer was established by injection of immunogen,which it was composed of the homogenate supernate of homogeneous oral mucosa from new zealand rabbits and Freund‘s complete adjuvant(FCA),into rabbit backs subcutaneously once every two weeks for 5 times,and the only FCA injected as normal control.Dihydromyricetin(37.5,75,150 mg·kg-1) and cydiodine(1 mg·kg-1) were daubed over oral mucosa in oral ulcer rabbits with syringe through blunt needle(16#) on d 7 after the last immunogen injection for 4 days,respectively.Water was given instead of drugs in normal and model control groups.The time of oral ulcer occurrence was observed and recorded everyday until the last administration of drugs.The titer of anti-oral mucosa antibody in sera was detected by turbidimetry,the level of IgG in sera was measured by ELISA and the pathological changes of oral mucosa were determined under the microscope the day after the last administration of drugs.Result: All of the rabbits in model occurred oral ulcers after 3 times of immunogen injection.The time occurred oral ulcers was 11.8±1.4(n=40) before drug‘s administration.In the model control group,the titer of antibody and the level of IgG in sera raised significantly,stratum corneum exfoliated,massive inflammatory cell infiltrated in lamina basal cell and propria of mucosa in oral ulcer sites,and score of pathological changes was 2.06±0.85.Dihydromyricetin(37.5,75,150 mg·kg-1) markedly decreased the times of ulcer occurrence,slightly lowered the titer of antibody to homogeneous oral mucosa and the level of IgG,and remarkably lightened the pathological changes of oral mucosa in occurred ucler sites.The scores of pathological changes were 1.81±0.91,1.37±0.80 and 1.12±0.61,respectively.Conclusion: Dihydromyricetin can promote healing of recurrent oral ulcer induced by homogenate supernate of homogeneous oral mucosa of rabbits as a immunogen.
