摘 要 ：目的:研究海风藤对人类神经母细胞瘤细胞系列淀粉样前体蛋白(β-APP)基因表达的抑制作用。方法:应用Northern印迹杂交观察β APPmRNA的变化,应用细胞存活率、乳酸脱氢酶(LDH)释放、细胞集落观察海风藤对神经细胞的直接作用。结果:海风藤选择性的抑制β-APP基因表达。实验浓度内未发现细胞毒性作用。结论:β-APP与Alzheimer’s病(AD)的发病关系密切,海风藤选择性抑制β-APP基因表达为其防治AD提供了一定的依据。

Abstract:Objective:To study the inhibition of β-amyloid precursor protein(β-APP) gene expression by Haifengteng.Method:Northern Blot analysis was used to investigate the level of β-APP mRNA,and cell viability,LDH release and colony formation assay were used to study the effect of Hairengteng of neurons.Result:Haifengteng inhibited the expression of β-APP gene selectively without causing cell injuries.Conclusion:β-APP is closely related to Alzheimer‘s disease.The findings in this study could be a basis for Haifengteng to have thrapeutic potentials in treating Alzheimer‘s disease.
