Abstract:ThecrystalizationtechnologyofcyclosporineAwasstudiedbyusingsinglefactortestandor
thogonalexperimentdesign.Efectsofdiferentsolvents,solventratio,andtemperatureonthecrystali
zationwereinvestigated.Resultsrevealedthatthewatercontenthadimportantefectsonthepurityand
recoveryofcyclosporineAandcrystalizationtemperaturewasalsoanotherimportantfactor.Theoptimum
crystalizationconditionsofcyclosporineAweredeterminedasfolows:thecoolingingradientwas
applied;crystaltemperaturewas-5℃;solventrationwas2∶1(acetone∶water),feedingtimeofwater
was30min;andcrystalizationperiodwas3h.TheaverageHPLCpurityofcyclosporineAreached
9915%.Theaveragecrystalinerecoverywas877%.
